Results of a new study into autism treatment may help in developing safer and more targeted therapies in the future.
The study done by scientists at UCLA Health found that brain and behavioral effects linked to autism may be temporarily reversible in adults using a single dose of the immunosuppressive drug rapamycin.
What did they study?
The scientists discovered that inflammation during pregnancy can trigger autism-like changes in offspring, according to the study published in Nature Communications. Knowing this, they exposed pregnant mice to a stimulus that caused mild inflammation.
The inflammation caused their offspring to have persistent inflammation in both the brain and the rest of the body. This is tied to behaviors associated with autism, such as abnormal brain growth, seizures and heightened sensitivity to sensory input.
How did they reverse autism-like symptoms?
During the study, the researchers gave a single dose of rapamycin to the adult offspring with autism-like symptoms. Within two hours, they found it significantly improved brain signaling and behavioral symptoms.
Rapamycin caused the offspring’s unusually active neurons to calm down, and repetitive behaviors and sensory overresponsivity eased. It also caused brain regions that had not been communicating properly to start showing more typical patterns.
What is rapamycin, and how does it affect the brain?
Rapamycin is a prescription immunosuppressant and mTOR inhibitor. It is primarily used to prevent organ transplant rejection but has recently gained attention for anti-aging therapies and seizure treatments via mTOR pathway inhibition, according to the National Institutes of Health.
mTOR stands for “Mechanistic Target of Rapamycin.” It acts as the cell’s “master switch” for regulating cell division, growth, metabolism and survival.
To understand its rapid effects, scientists examined gene activity in brain cells before and after rapamycin treatment.
They concluded that rapamycin was changing brain function and altering its electrical activity, rather than rebuilding the brain’s underlying structure, which typically takes longer. This reversed abnormal patterns of gene expression tied to autism and epilepsy.
What does this mean?
The results point to several possible targets for future treatments that could potentially address common autism symptoms that are difficult to treat, including sensory overresponsivity.
“The level of functional normalization achieved over this short time suggests new mechanisms by which possible treatments may act,” said the study’s senior author, Dr. Harley Kornblum, in an interview with Science Daily. “It suggests the adult brain may be more adaptable than we assumed, even when the underlying structural changes from early development are still there.”
Why aren’t researchers calling rapamycin a treatment for autism?
While the results of the mouse study were promising, the benefits of rapamycin were only temporary, and the drug’s effects were too rapid to produce physical changes in the brain caused by inflammation.
Rapamycin is also toxic with repeated use, and the researchers found that the mice developed a tolerance to it after several weeks, meaning the benefits in the initial dose were far less prominent.
While there are no trials in humans any time soon, Dr. Neil Harris, the study’s co-senior author, told Science Daily the results alone are a jumping-off point for future therapies.
“This points toward new therapeutic targets like sensory circuit neuromodulation or balancing neuronal inhibition and excitation, rather than toward rapamycin itself as a treatment.”
